Asiatic acid exerts neuroprotective effect against hypoxicischemic brain injury in neonatal rats via inhibition of oxidative damage

Purpose: To investigate the effect of asiatic acid on hypoxic ischemia-induced injury in neonatal rats, and underlying mechanism action.Methods: Hypoxic-ischemia (HI) rat model was established via permanent ligation carotid artery, followed by hypoxia (exposure to 8 % oxygen 92 nitrogen) for 24 h. Immunofluorescence, using fluorescence microscope, used determination expressions p-TAK1, NeuN GFAP. Western blotting assaying protein expression levels, while TUNEL assay employed measurement apoptosis.Results: Treatment rats with prior HI effectively prevented up-regulation pTAK1 decreased count p-TAK1-containing astrocytes. The proportion containing p-TAK1 brain cortex significantly reduced (p < 0.05). suppressed HI- induced pJNK expression. HI-induced increase levels caspase-3, p53 p-c-Jun were reversed HImediated p- JNK, inhibited NG25. Asiatic treatment also alleviated HI-mediated apoptosis neurons cortex, when compared group 0.05).Conclusion: These findings suggest that prevents inhibition neuronal apoptosis. Moreover, it inhibits TAK1 activation, suppresses p-JNK targets pro-apoptotic factors cortex. Therefore, may be a therapeutic agent management injury.
